Are biopsies always performed during a colposcopy?

No, they are not. In some cases the doctor will decide that they are not necessary.

In other cases, when the lesions found are very serious and cover a large area, the doctor knows that they cannot count on isolated biopsies and that they must surgically remove a larger area in order to exclude invasive cancer (which may not be visible during the colposcopy).
